    Promoting Computer Science with Video Games: Teaching Object-Oriented Programming through Unity

    In recent years, the demand for people educated in computer science has continued to grow as the possible applications of software expand. For this reason, it is important to not only offer young adults the ability to learn computer science, but also to intrigue them with interesting and engaging applications of computer science. Organizations, such as the CollegeBoard and Exploring Computer Science, have been working to make computer science education more available and desirable to younger audiences. This project contributes to these goals by creating and implementing a curriculum for teaching object-oriented programming to high school students through video game programming. Students were taught how to use a third-party game engine called Unity to create complex 3D games. The curriculum was designed to provide students with a basic understanding of the tools and concepts they needed to make a game, and provided students with several hours of learning time to create their own games. This course was taught at San Luis Obispo High School to a group of 20 students. A survey was taken by 14 of the 20 students before and after the course. This survey revealed that half of the students who did not understand objects in programming before the course did understand objects after the course. The survey also showed that many students were able to learn C#, an object oriented programming language, during the course. Lastly, most of the students ended the course with a completed project that could be added to a portfolio

    Interaction of Oral Bacteria With Gingival Epithelial Cell Multilayers

    Primary gingival epithelial cells were cultured in multilayers as a model for the study of interactions with oral bacteria associated with health and periodontal disease. Multilayers maintained at an air-liquid interface in low calcium medium displayed differentiation and cytokeratin properties characteristic of junctional epithelium. Multilayers were infected with fluorescently labeled Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum or Streptococcus gordonii, and bacterial association was determined by confocal microscopy and quantitative image analysis. P. gingivalis invaded intracellularly and spread cell to cell. A. actinomycetemcomitans and F. nucleatum remained extracellular and showed intercellular movement through the multilayer. S. gordonii remained extracellular and predominantly associated with the superficial cell layer. None of the bacterial species disrupted barrier function as measured by transepithelial electrical resistance. P. gingivalis did not elicit secretion of proinflammatory cytokines. However, A. actinomycetemcomitans and S. gordonii induced IL-1ÎČ, TNF-α, IL-6 and IL-8 secretion; and F. nucleatum stimulated production of IL-1ÎČ and TNF-α. A. actinomycetemcomitans, F. nucleatum and S. gordonii, but not P. gingivalis, increased levels of apoptosis after 24 h infection. The results indicate that the organisms with pathogenic potential were able to traverse the epithelium, while the commensal bacteria did not.     FLASH Irradiation Results in Reduced Severe Skin Toxicity Compared to Conventional-Dose-Rate Irradiation

    Radiation therapy, along with surgery and chemotherapy, is one of the main treatments for cancer. While radiotherapy is highly effective in the treatment of localized tumors, its main limitation is its toxicity to normal tissue. Previous preclinical studies have reported that ultra-high dose-rate (FLASH) irradiation results in reduced toxicity to normal tissues while controlling tumor growth to a similar extent relative to conventional-dose-rate (CONV) irradiation. To our knowledge this is the first report of a dose-response study in mice comparing the effect of FLASH irradiation vs. CONV irradiation on skin toxicity. We found that FLASH irradiation results in both a lower incidence and lower severity of skin ulceration than CONV irradiation 8 weeks after single-fraction hemithoracic irradiation at high doses (30 and 40 Gy). Survival was also higher after FLASH hemithoracic irradiation (median survival >180 days at doses of 30 and 40 Gy) compared to CONV irradiation (median survival 100 and 52 days at 30 and 40 Gy, respectively). No ulceration was observed at doses 20 Gy or below in either FLASH or CONV. These results suggest a shifting of the dose-response curve for radiation-induced skin ulceration to the right for FLASH, compared to CONV irradiation, suggesting the potential for an enhanced therapeutic index for radiation therapy of cancer

    TRY plant trait database – enhanced coverage and open access

    Plant traits - the morphological, anatomical, physiological, biochemical and phenological characteristics of plants - determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait‐based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits - almost complete coverage for ‘plant growth form’. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait–environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    RE-EXAMINING THE EVIDENCE FOR LATE COLONIZATION ON EASTER ISLAND

    One of the recent discussions to emerge among archaeologists regarding Rapa Nui (Easter Island) prehistory contrasts "early" and "late" estimates for initial human colonization of the island. These differing estimates, in tum, offer significantly different messages for the timing and rate of cultural evolution on the island. A recent study of eleven charcoal samples concluded that Rapa Nui was first colonized around 1200 CE. A new analysis of the same eleven charcoal samples suggests that the data are consistent with an earlier colonization date, around 900 CE. The three hundred year difference between the two estimates could mean the difference between a "short chronology" and "long chronology" to archaeologists and environmentalists alike.</p

    Effect of conspecifics and crayfish on zebra mussel defense behavior.

    Zebra mussels (Dreissena polymorpha Pallas, 1769) have profoundly negative effects on both Great Lakes aquatic ecosystems through the decline of native bivalve species and on economic activities, costing industry upwards of $150,000,000 yearly. Successful control of these invasive species would be beneficial for the Great Lakes region as a whole. Recently, studies have shown that zebra mussels respond to both injured mussel conspecifics and to predator kairomones, but there has been no conclusive evidence to suggest that either has more of an effect than does the other. In a laboratory setting, we tested this question using four treatments: a control tank, a tank containing crushed mussel conspecifics, a tank containing crayfish that had eaten mussels, and a tank containing crayfish exclusively. Testing the distance that mussels traveled as well as the aggregation rates of mussels in each treatment tank, we determined that mussels move significantly farther when exposed to crushed mussel conspecifics than in the absence of any treatment. While not significant, trends show that crushed conspecifics have the greatest effect on mussel clumping rates as well. This signifies that the presence of injured mussels triggers greater behavioral changes in zebra mussel populations than does the presence of predator kairomones.     The use of modelling studies to inform planning of health services: case study of rapidly increasing endoscopy services in Australia

    Background Demand for gastrointestinal endoscopy in Australia is increasing as a result of the expanding national bowel cancer screening program and a growing, ageing population. More services are required to meet demand and ensure patients are seen within clinically recommended timeframes. Methods A discrete event simulation model was developed to project endoscopy waiting list outcomes for two large metropolitan health services encompassing 8 public hospitals in Australia. The model applied routinely collected health service data to forecast the impacts of future endoscopic demand over 5 years and to identify the level of service activity required to address patient waiting times and meet key policy targets. The approach incorporated evidence from the literature to produce estimates of cost-effectiveness by showing longer term costs and Quality Adjusted Life Years (QALYs) associated with service expansion. Results The modelling revealed that doing nothing would lead to the number of patients waiting longer than clinically recommended doubling across each health service within 5 years. A 38% overall increase in the number of monthly procedures available was required to meet and maintain a target of 95-98% of patients being seen within clinically recommended timeframes to the year 2021. This was projected to cost the funder approximately 140millioninadditionalactivityovera5yearperiod.Duetoimprovedpatientoutcomesassociatedwithtimelyintervention,itwasestimatedthattheincreasedactivitywouldgenerateover22,000additionalQALYsacrossthetwohealthservices.Thistranslatedtoanincrementalcost−effectivenessratioof140 million in additional activity over a 5 year period. Due to improved patient outcomes associated with timely intervention, it was estimated that the increased activity would generate over 22,000 additional QALYs across the two health services. This translated to an incremental cost-effectiveness ratio of 6467 and $5974 per QALY for each health service respectively. Conclusions Discrete event simulation modelling provided a rational, data based approach that allowed decision makers to quantify the future demand for endoscopy services and identify cost-effective strategies to meet community needs
